pharmacological studies on khamira marwarid: effect on pentylenetetrazole-induced seizures, cognition and biochemical markers in mice

چکیده
cognitive impairment in epileptics may be a consequence of the epileptogenic process as well as antiepileptic medication. thus, there is need for drugs, which can suppress epileptogenesis as well as prevent cognitive impairment. in the present study, the effect of khamira marwarid (kah 1 ), a formulation based on indian system of unani medicine, was evaluated on the course of pentylenetetrazole (ptz)-induced seizures, learning deficit and oxidative stress markers in mice. male albino mice were injected ptz (65 mg/kg sc) on the 5 th day of the treatment for the development of seizures. spontaneous alternation behaviour was carried out on the 1 st and the 5 th day of the treatment after ptz administration, while the oxidative stress parameters (malondialdehyde and glutathione) were measured in the whole brain upon the completion of the behavioural assessment. the administration of khamira marwarid (kah 1 ), 50 mg/kg significantly decreased the ptz-induced seizures and showed improvement in the learning deficit induced by ptz as evidenced by the increased latency time and frequency of jerks and improvement in sab. the findings suggest the potential of khamira marwarid (kah 1 ) as adjuvant to antiepileptic drugs with an added advantage of preventing cognitive impairment.

متن کاملArsenic and Oxidative Stress in Pentylenetetrazole-induced Seizures in Mice
Background and Objective: Chronic arsenic toxicity is a widespread problem; the role of brain oxidative stress has been suggested in the genesis of epilepsy and in the post-seizure neuronal death. However, studies investigating the effects of arsenic on seizure and related mechanisms are limited. The purpose of this study was to examine the effect of prolonged exposure to sodium arsenite on oxi...
متن کاملEvaluation of Anticonvulsant Effects of Nimodipine and Ascorbic Acid on Pentylenetetrazole- Induced Seizures in Mice
Introduction: The mechanisms underlying the vulnerability of the brain to seizures remains unknown. Calcium ions influx and oxidative stress have been implicated in a variety of acute and chronic epileptogenic conditions. The present study was aimed at investigating the effects of nimodipine and ascorbic acid (vitamin C) alone and in combination on pentylenetetrazole (PTZ) - induced seizures ...
متن کاملInhibitory and Dose-dependent effect of intracerebroventricular docosahexaenoic acid on clonic seizures induced by pentylenetetrazole in mice
Introduction: Epilepsy is one of the most common neurologic disorders. Pharmacoresistance and adverse effects of current antiepileptic drugs (AEDs) necessitate development of new drugs and strategies for treatment of epilepsy. Omega 3-Polyunsaturated fatty acids (ω3-PUFAs) are safe nutritional supplements that recently considered for treatment of epilepsy. Anticonvulsant effect of docosahexa...
متن کاملAnticonvulsant Effect of Alcea aucheri on Pentylenetetrazole and Maximal Electroshock Seizures in Mice
Introduction: This study was designed to investigate the possible anticonvulsant effect of acute administration of an aqueous extract of flowers of Alcea aucheri (EFA) in a seizure model. Methods: Seizures were induced in male adult Swiss mice by administration of pentylenetetrazol (PTZ) or Maximal Electroshock (MES). Mice were randomly subjected to receive saline, EFA (8.75-175 mg.kg-1), or d...
متن کاملEffect of Ocimum basilicum hydro-alcoholic extract on oxidative damage of brain tissue following seizures induced by pentylenetetrazole in mice
Introduction: A relationship between epileptic seizures and brain tissue oxidative damage has been suggested. Ocimum basilicum (O. basilicum) has been shown to have beneficial effects including hypnotic and protective against tissue oxidative damage. The present study was designed to evaluate the effects of O. basilicum hydro-alcoholic extract on oxidative damage of brain tissue following s...
